Abstract: Hematopoiesis requires highly coordinated gene expression through transcriptional and post-transcriptional mechanisms. By comparing enhancer landscape, transcriptional factor occupancy, 3D chromatin structure and transcriptomic changes during blood cell differentiation, I identified lineage-specific enhancers and macro-molecular interactions driving enhancer activation, providing initial insights into enhancer dynamics and gene expression programs directing lineage-specific transcription during hematopoiesis. Further comparison between the proteomic and transcriptomic changes in erythroid development uncovered post-transcriptional pathways leading to enhanced mitochondrial biogenesis. Perturbation of mitochondria interrupted intracellular metabolism, leading to altered transcriptional regulation and defective lineage differentiation, which provide a new pathway between mitochondrial biogenesis and epigenetic gene regulation indispensable for normal erythropoiesis.
